The new supermarket - which employs 102 people - will open its doors to shoppers at 9am Wednesday, September 16.
The supermarket forms part of LWP's $1.5 billion, master-planned community near Branxton.
Huntlee will eventually be home to 20,000 residents in 7500 dwellings, with a 200-hectare town centre.
Huntlee Shopping Centre will be the first retail centre to open in the new town.
Coles Huntlee store manager Tim Hill said he was delighted with the new store and the opportunity to support local jobs.
"We're excited to be launching a brand-new supermarket for people in the Hunter Valley. The modern store brings all the latest customer offerings from the big city right to the people of Huntlee," Mr Hill said.
"We're so proud of the hard work everyone has put in to get the store launch ready.
"It's fantastic that we're creating more than 100 local jobs on opening day - we're certainly going to be a big employer the Hunter Valley.
"We really want to take a personal approach and get to know our customers so we become part of the community.
"I'm looking forward to seeing what we can achieve in the next 12 months and over the years to come."
LWP project director Stephen Thompson welcomed Coles and Liquorland to Huntlee and said he cannot wait to see the locals explore the new shopping centre.
"It will be a great day for the residents of Huntlee and those living in surrounding villages, who will reap the benefits of the new local Coles and Liqourland stores, making a huge difference to their day-to-day lifestyle," he said.
"The opening of the Huntlee Shopping Centre has been highly anticipated by local residents, and to have an offering of this calibre on their doorstep is sure to delight many.
"It's only fitting we welcome Coles as the first major retail project in the new 200-hectare town centre which, over the coming years, will evolve into a major regional hub."
Click and Collect will also be available at the store, allowing shoppers to order their groceries online and have them delivered to their car boot.
Coles Huntlee will officially open to shoppers at 9am Wednesday, September 16 and will be open seven days a week at 22 Empire Street, Branxton.
