The proposed site at 217-219 Vincent Street is currently home to Eaton's Mitre 10, which is moving to new premises further south on the main street.
If approved, it would be the fast food giant's second restaurant in Cessnock, with the company intending to keep the Allandale Road site open.
McDonalds will apply for the Vincent Street restaurant to trade 24 hours a day, seven days a week when it lodges plans with Cessnock City Council following completion of a community survey, which closes next Sunday.
A McDonalds Australia spokesperson said the proposal is in response to "growing demand for food and beverages in the area".
"The proposed development would provide the opportunity for significant financial investment into the local area via job creation and construction of the site," the spokesperson said.
"If approved, it would inject approximately $5 million into the local economy and create approximately 100 jobs during construction phase, as well as a further 100-to-120 jobs once operational."
McDonalds is conducting an online survey before it lodges a development application with Cessnock City Council, to investigate potential concerns and impacts, as well as benefits and opportunities that may result from the development.
The survey closes Sunday, December 20 and can be completed at bit.ly/vincentstreetmcdonalds.
