The challenge runs from May 15 to 22, and the club's team hopes to run 650 kilometres and raise $6500 for the charity.
"Sadly, 65,000 Australians attempt to take their life each year," club manager Adeshia Gillies said.
"That's why Anytime Fitness Cessnock is taking action to raise funds and awareness for R U OK.
"We meet so many people here every day who struggle with their mental health.
"Creating awareness is really important, as is giving people a safe space to speak up."
As part of its fundraising activities, the club will host a community day at Cessnock Sportsground this Saturday from 7.30am to 10am.
Activities will include a boot camp for all ages, kids' games, a face painter, firefighting demonstrations, and a breakfast barbecue supplied by the Rotary Club of Cessnock.
The team from headspace will also be on hand to raise awareness of their service.
Entry is $5 per person, and bookings can be made at https://bit.ly/3eygiMH.
Donations can also be made online at treadasone.com.au.
